Childs, Platoon 2078, Company E, a 19-year-old native of Springdale, Ark., attributes his success in recruit training to preparing for it an entire year before shipping to Marine Corps Recruit Depot, San Diego. He was prepared to go into recruit training, said Denton Childs, his father. He worked out every day, read books, and knew what to expect. In addition to knowing what he was getting into, Childs knew why he was getting into it. He always wanted to be the best of the best, and that is why he chose the Marine Corps over the other services, according to Denton. In everything I do, I want to do my best, said Childs. That is why I didnt join the Navy or the Army. He wanted as much of a challenge as he could get, said Denton. With the challenge of recruit training in his sights, Childs knew the Marine Corps wouldnt be easy, according to Nereida Childs, his mother. He worked out, bu ilding himself physically and made a healthy diet high on his list of priorities. His recruiters and fellow poolees in the Delayed Entry Program gave him the name Turbo for the amount of crunches he could pump out at the initial strength tests. According to Childs, he could do about 110-120 crunches in two minutes compared to his 167 crunches now. The positive physical payoff of Childs hard work prior to and during recruit training can be seen in his increase from 23 pull-ups to 29 pull-ups and the decrease in his three-mile run time from 22 minutes to 20 minutes. Childs felt he was a weak swimmer, but applied his be the best attitude and ended up becoming a first-class swimmer, one of only three out of his 54-man platoon. Childs excelled on the rifle sortiment as well, scoring 237 out of 250 qualifying as an expert rifleman, even though he had never shot a rifle before. He always worked harder than everyone in sports, said his father. He did it harder and got better than everyone. On top of always striving to be the best, Childs has always helped others who were falling behind and stuck up for the underdog, according to his mother. It was displayed in recruit training by his ability to make his platoon work together to get the missions accomplished. I was put as guide for my pull-ups, and then I grew from there, he said. I learned things and made the recruits work as a team. Childs said he displays his leadership through his bearing and the example he sets. I believe in giving 100 percent at all times, he said. Even though it is not always easy. It is a continuous cycle for this new Marine to look for tough challenges and complete them before moving on to the next. I give until I know I cant get any better or give more, said Childs. Childs said he dreams of living in San Diego on the beach with financial stability and a beautiful wife. People always assumed that cartilage would be an easy tissue to grow because it is very simple, consisting of only one type of cell, no blood vessels and no nerves, says Dr. Gordana Vunjak-Novakovic, the Mikati Foundation Professor of Biomedical Engineering at Columbia Engineering who led the study. However, although the cartilage that scientists grew in the past looked and felt like cartilage, the mechanical properties were not there. Two mechanical properties of cartilage are essential mechanical stiffness and lubrication, Vunjak-Novakovic explains. If you compress the cartilage, it should resist the force because thats how cartilage resists force in our joints. If y ou slide another piece of tissue over cartilage, it should have very low friction. The measurements for stiffness and friction never achieved the normal range of human cartilage, she says.Until now, researchers have had success in engineering pieces of cartilage using young animal cells and also in creating synthetic replacement cartilage in the lab. This breakthrough could help millions of people who suffer pain or lack of mobility as cartilage cushioning joints erodes over time or is damaged by injury. Vunjak-Novakovic says both her gruppes approach and the synthetic approach developed by researchers at Duke Un iversity have good potential for clinical application. Certainly, we do not have a definitive solution, and there may not be a single solution for all patients and clinical situations, she adds.About a year ago, stumped after years of work, members of Vunjak-Novakovics team decided to forget everything we had tried, look back at biology books, and see how the body makes cartilage, she says. Our overall approach in the lab is to design our tissues based on biological principles. Everyone was always starting from the step when cartilage has been made in the body so we decided maybe there is something in the previous step that is priming the cells to do their job differently.In the body, the cells that form the cartilage first form little cell bodies in which cells are tightly packed. This triggers some of the biological processes that give rise to cartilage. This was never introduced in vitro before,says Vunjak-Novakovic. We did that and continued to culture the forming tissue for sev eral weeks. At the end of the process we had this very beautiful stratified structure. We achieved for the first time complete stiffness and tribology of cartilage.It is especially rewarding, she says, because this method is extremely simple and something everyone can do.Others involved in the project include Dr. Sarindr Bhumiratana, postdoctoral fellow in Vunjak-Novakovics lab and the leading author of a paper describing the study Dr. Gerard Ateshian, chair of the mechanical engineering department and professor of mechanical engineering and biomedical engineering and his PhD student, Sevan Oungoulian, who helped perform some of the measurements. Asked why it took so long to achieve, Vunjak-Novakovic says, Its interesting how in life you often say Why didnt I think about this earlier? Its exactly this. According to her, one of the next steps is using the model in a lab to further some of the developmental processes and study them in the context of disease, for example, modeling oste oarthritis in order to understand the progression of the disease and to look for drugs or measures that would be helpful. Another is to conduct animal studies to determine the durability of the cartilage and whether it develops further in the body to be able to predict how it would work in human patients.As the project moves forward, 3D printing may play an important role in multiplying the mesenchymal cells isolated from bone marrow or fat to produce the very small, uniformly sized cell bodies that generate the functional human cartilage tissue. We used a very simple method for making the cell bodies, but one can envision that clinical reconstruction of large surfaces of cartilage lost to injury or disease would require fast 3D printing of large numbers of cell bodies directly onto anatomically shaped pieces of engineered bone to grow a graft for implantation, says Vunjak-Novakovic.This is teamwork, she points out.
